Mutational analysis of the Drosophila DNA repair and recombination gene mei-9.

چکیده
Drosophila mei-9 is essential for several DNA repair and recombination pathways, including nucleotide excision repair (NER), interstrand crosslink repair, and meiotic recombination. To better understand the role of MEI-9 in these processes, we characterized 10 unique mutant alleles of mei-9. These include a P-element insertion that disrupts repair functions but not the meiotic function; three nonsense mutations, one of which has nearly wild-type levels of protein; three missense mutations, one of which disrupts the meiotic function but not repair functions; two small in-frame deletions; and one frameshift.

Heteroduplex DNA in meiotic recombination in Drosophila mei-9 mutants.
Meiotic recombination gives rise to crossovers, which are required in most organisms for the faithful segregation of homologous chromosomes during meiotic cell division. Characterization of crossover-defective mutants has contributed much to our understanding of the molecular mechanism of crossover formation. We report here a molecular analysis of recombination in a Drosophila melanogaster cros...
متن کاملDrosophila MUS312 and the vertebrate ortholog BTBD12 interact with DNA structure-specific endonucleases in DNA repair and recombination.
DNA recombination and repair pathways require structure-specific endonucleases to process DNA structures that include forks, flaps, and Holliday junctions. Previously, we determined that the Drosophila MEI-9-ERCC1 endonuclease interacts with the MUS312 protein to produce meiotic crossovers, and that MUS312 has a MEI-9-independent role in interstrand crosslink (ICL) repair. The importance of MUS...

Meiotic recombination and DNA repair are mediated by overlapping sets of genes. In the yeast Saccharomyces cereuisiae, many genes required to repair DNA doublestrand breaks are also required for meiotic recombination. In contrast, mutations in genes required for nucleotide excision repair (NER) have no detectable effects on meiotic recombination in S. cereuisiae. The Drosophila melanogaster mei...
متن کاملGenotoxic effects of strong static magnetic fields in DNA-repair defective mutants of Drosophila melanogaster.
To assess the possibility that strong static magnetic fields cause DNA damage and mutation, we examined the genotoxic effects of magnetic field exposure by using the somatic mutation and recombination test system in DNA repair-proficient and -deficient strains of Drosophila melanogaster. A postreplication repair-defective mutation mei-41D5 and/or a nucleotide excision repair-defective mutation ...
متن کاملCyclin G is involved in meiotic recombination repair in Drosophila melanogaster.
Cyclin G (CycG) belongs to the atypical cyclins, which have diverse cellular functions. The two mammalian CycG genes, CycG1 and CycG2, regulate the cell cycle in response to cell stress. Detailed analyses of the role of the single Drosophila cycG gene have been hampered by the lack of a mutant. We generated a null mutant in the Drosophila cycG gene that is female sterile and produces ventralise...
